  • Description: HTTase is a water soluble protein that can covalently conjugate virtually any protein of interest with a variety of small molecules in less than 60 minutes. Conjugation by HTTase is residue specific and is active in physiological buffer at room temperature. This one-pot method for bi-molecular conjugation differs from existing techniques in that only the modifier substrate needs to be added to the "pot". Since the HTTase catalyst is fused to the protein substrate the need for cofactors or accessory proteins is eliminated. Only a single glycine residue, added to the protein target, is required for conjugation. Virtually traceless labeling is a key advantage of this system, which differentiates it from existing protein conjugation methods. Additional features include; two component labeling kinetics, site-specific, stoichiometric modifications and broad substrate tolerance.
